The Universal V1-Puck (Unipuck) is a sample pin storage and shipping container compatible with many automated sample mounting systems. The V1-puck holds 16 MiTeGen sample bases/pins. It consists of two parts shown in the image: 1) the sample enclosure (left) and 2) the sample base piece (right).

Each part of the V1-puck has a unique serial number for identification. Custom serial numbers are available if requested at time of order.

    The Universal Puck Project

    A collaboration to develop the uni-puck, a new standard sample pin storage and shipping container.

    The uni-puck has been designed to be compatible with all robotic mounting systems currently in use at synchrotrons in the United States and many systems used abroad, requiring minimal changes to the robotic hardware of each system.

    Introduction

    The availability of robotic sample mounting systems continues to increase at synchrotron sources. (See smb.slac.stanford.edu/robosyncfor a list of available devices). Although the majority of these systems are compatible with Hampton-style magnetic cryo-pins, many types of containers are used for sample pin transportation and storage.

    Currently at synchrotron stations in the United States there are three main sample storage containers used: the ALS puck, the SSRL cassette, and the MSC magazine. The costs to purchase these containers and associated cryo-tools and the effort required to master their use has become a significant impediment to groups seeking to take advantage of more than one of these facilities.

    In an effort to minimize this burden, a collaboration was formed between the ALS, APS SBC-CAT, and SSRL staff. This group first proposed construction of a sorting system that would transfer sample pins between the most commonly used containers. However, it was observed that it could be possible to develop a sample storage container that would be compatible with all the sample mounting systems currently in use in the United States and many of the systems used abroad. Therefore, during a meeting at the 2004 ACA conference in Chicago the group decided to develop a universal container. We have received a lot of positive feedback in response to this effort. In particular, many have joined this collaboration including developers at the NSLS, APS, and CHESS and of commercial systems. These developers were asked to test the V1 uni-puck with their automated mounting systems and provide feedback on the design with the goal of cross compatibility and reliability. Through the collaborative efforts of all involved, a first version of the universal container, the V1 uni-puck has been completed. The V1 uni-puck was released for use at the ALS sector 5.0 beam lines and at the SSRL macromolecular crystallography beam lines with test users. Once groups have more experience using the V1 uni-puck, a second version (V2) may be released if necessary for improved performance.

    Design

    The outside form factor of the new universal container (uni-puck), resembles a standard ALS puck and for this reason will be compatible with the standard ALS cryo-tools and fit inside the ALS shipping dewar canister. Unlike the ALS puck, the pins are held inside the container using rare earth magnets which are more durable and more strongly and reliably hold the pins in place. Magnets are used in both the enclosure piece and base piece of the uni-puck. The uni-puck enclosure resembles an SSRL cassette as it contains ports that are nearly identical to those in an SSRL cassette. The uni-puck also provides the same clearance around the sides and bottom of the sample pin as an MSC magazine and the base piece shares a similar hole and slot for alignment inside the dispensing dewar.

    The universal puck consists of two parts: 1) the base piece and 2) the sample enclosure. Some key differences between the original ALS puck and universal puck are illustrated in the figures below.

    Both the universal puck enclosure and the base contain magnets and the assembly will hold together by magnetic force. Therefore, the side clips used on the ALS puck for this purpose are not necessary. However to enable the universal puck parts to fit interchangeably with ALS puck parts, clearance and mounting holes for these clips are provided. Interchangeability of the universal puck base and enclosure with the ALS puck parts enable rapid transfer of samples between these two containers.
    Compatibility of Specific Robotic Systems

    The ALS Robot (Berkeley Automated Mounting system)

    The uni-puck base piece is used inside the ALS dispensing dewar. This universal base will work in the ALS robotic systems interchangeably with the original ALS puck base. Because the sample pins are held in the same position, minimal hardware modifications or reprogramming is required.

    Because the outside form factor is similar, the V1 uni-puck is compatible with the standard ALS cryo-tools and shipping dewar canister. Additionally, the uni-puck base and enclosure pieces fit on the original ALS puck enabling rapid transfer between the two types of containers.

    Members of the ALS beam line development group are taking part in the universal puck project and will ensure compatibility and reliability with their automounter.

     

    The Stanford Automated Mounting System (SAM)

    The enclosure piece of the universal puck is used inside the SAM dispensing dewar. This piece contains magnetic cavities nearly identical to those in a SSRL cassette. To adapt the enclosure piece to the standard SAM setup an adaptor cassette has been developed. Four universal pucks may be inserted into one adaptor cassette. The adaptor is then inserted inside the robot dispensing dewar using the cassette transport handle (see below).

    The original SSRL cassettes will continue to work with the SAM system. Within the SSRL dispensing dewar the 3 cassette locations are each capable of interchangeably holding an SSRL cassette or an adaptor cassette containing 4 universal pucks.

    Once the universal container project and adaptor cassette development is complete, SSRL users will have the option to use cassettes or uni-pucks and the SAM system will be programmed to sort samples between these two options.
    APS SBC-CAT Automatic Mounting System

    Currently the SBC-CAT system works with MSC magazines. To work with the uni-puck, the system will be reprogrammed and a new dewar base plate will be developed that can accommodate both MSC magazines and universal puck bases. Users will then have the option to use either container.

    For added reliability when picking up a sample pin, the SBC-CAT robot grippers surround the bottom edge of the pin trapping it in place (rather than gripping the side of the pin and relying on friction to hold it). To accommodate this gripper, the uni-puck base provides additional clearance around the bottom and side of the pin base. This clearance is equal to that provided by the MSC magazine.

    Members of the APS SBC-CAT development group have been instrumental in the universal puck project and will ensure compatibility and reliability with their automounter.

    Commercially Available Systems

    This universal container concept has received positive feedback from the commercial manufacturers of robotic sample mounting systems (MSC, Bruker, and Mar). Moreover, MSC and Bruker representatives feel that it would be rather straight-forward for them to offer an alternative dewar base plate and programming to accommodate the proposed universal puck design.

    Fabrication of an alternative enclosure, that would contain traditional cryo-vials and mate with the universal puck base, will make it easy to transfer samples from vials into pucks and enable compatibility with even more robotic mounting systems.

DESCRIPTION

Siliconized glass coverslips allow a droplet to be suspended in a position which provides near optimal conditions for vapor diffusion with the surrounding reservoir solution. These siliconized glass cover slides provide a consistent, high quality finish for crystallization experiments. The hydrophobic surface produces a drop which “stands well” and does not flatten on the glass. The siliconized surface prevents the adhesion of crystals and precipitate onto the glass surface. Use vacuum grease or oil to seal cover slide to plate. Available in 18 or 22 mm diameter circles.

DESCRIPTION

The CrystalCap ALS is a magnetic sample mount (also known as a cap, pin or goniometer base) designed for cryocrystallography systems that accept ALS style sample mounts. The CrystalCap ALS attaches to a magnetic CrystalCap Vial and magnetic goniometer base. The tip of the CrystalCap accepts a Mounted CryoLoop™.

CrystalCap ALS (HR4-779) is supplied without a vial (HR4-904).

The CrystalCap Vial is a 1.8 ml cryo vial featuring two small vents. A ring magnet is molded into the open end of the vial so that when the cap is positioned in the vial, the ring magnet holds the cap on the vial during cryogenic storage. The HR4-904 CrystalCap Vial does not have a magnet on the bottom of the vial.

The ledge free conical shape of the CrystalCap ALS is compatible with ALS style grippers, auto mounters and sample handlers.

描述

Adhesive, malleable, non-hardening clays have many uses in the crystallization lab. Description 
Similar to the Mounted CryoLoop™ in design and function, the Adjustable Mounted CryoLoop allows one to manipulate and orient the CryoLoop to multiple, unique positions previously unattainable with typical cryo setups. The Adjustable Mounted CryoLoop is a 20 micron diameter synthetic loop mounted inside a malleable stainless steel sleeve, which in turn, is mounted inside the EasySnap MicroTube™.

Using forceps or a probe one can manipulate the angle and orientation of the CryoLoop by bending and adjusting the angle of the malleable stainless steel insert which holds the CryoLoop. The Adjustable Mounted CryoLoop may be positioned several times. Note: Repeated manipulation will fatigue the sleeve which can lead to failure of the sleeve. The Adjustable Mounted CryoLoop is designed to work with the CrystalCap System components.

Description 
The CrystalCap ALS is a magnetic sample mount (also known as a cap, pin or goniometer base) designed for cryocrystallography systems that accept ALS style sample mounts. The CrystalCap ALS attaches to a magnetic CrystalCap Vial and magnetic goniometer base. The tip of the CrystalCap accepts a Mounted CryoLoop™ or MicroTube™ fitted with a CryoLoop™.

CrystalCap ALS (HR4-779) is supplied without a vial (HR4-904).

The CrystalCap Vial is a 1.8 ml cryo vial featuring two small vents. A ring magnet is molded into the open end of the vial so that when the cap is positioned in the vial, the ring magnet holds the cap on the vial during cryogenic storage. The HR4-904 CrystalCap Vial does not have a magnet on the bottom of the vial.

The ledge free conical shape of the CrystalCap ALS is compatible with ALS style grippers, auto mounters and sample handlers.

ALS style sample mounters accept the CrystalCap ALS.

Description
The CrystalCap SPINE HT is a magnetic sample mount (also known as a cap, pin or goniometer base) designed for cryocrystallography systems that accept SPINE style caps. The CrystalCap SPINE HT attaches to a magnetic CrystalCap SPINE Vial and magnetic goniometer base. The tip of the CrystalCap SPINE HT accepts a Mounted CryoLoop™ or MicroTube™ fitted with a CryoLoop™.

The CrystalCap SPINE Vial is a 1.8 ml cryo vial featuring two small vents and is compatible with the CrystalCap SPINE HT. A ring magnet is molded into the open end of the vial so that when the cap is positioned in the vial, the ring magnet holds the cap on the vial during cryogenic storage. The CrystalCap SPINE Vial features chamfered edges for enhanced cap positioning as well as a magnetic alloy bottom for stability. The HR8-114 CrystalCap SPINE Vial does have a magnet on the bottom of the vial.

The hollowed out design of the CrystalCap SPINE HT is compatible with SPINE style grippers, auto mounters and sample handlers.

The CrystalCap SPINE HT features a two dimensional (2D) alphanumeric 16 x 16 data matrix code on the underside of the cap. Each cap is also color coded for CryoLoop size.

Description
The CrystalCap ALS HT is a magnetic sample mount (also known as a cap, pin or goniometer base) designed for cryocrystallography systems that accept ALS style sample mounts. DESCRIPTION  描述

Crystal Clear Sealing Film is an optically transparent sealing film for SBS format 24, 48, 96 and 384 well plates. The 2 mil +/-10% (0.05 mm) thick, optically clear, non-fluorescing polyester film with 1.0 – 1.3 mil (0.025 mm) thick acrylic custom adhesive is designed for the convenient and secure sealing of crystallization plates. A split, optically opaque 2.0 mil +/-10% polyester backing with two end tabs assures a clean uniform adhesive layer and allows for easy, accurate positioning on the plate. Before use, peel off the opaque center protective polyester backing to reveal the optically clear sealing film. Film length: 5.625″ +/- 0.015 Film width: 3.125″ +/- 0.015 End tabs: 0.375″ +/- 0.015 Perforations: 0.415″ +/- 0.015 from edge of film Corner radius: 3/32″. Free of detectable levels of DNase-, RNase- and Human/Mouse DNA contamination (PCR Certified), non-sterile. Recommended temperatures from -40 to +120°C. Not recommended for UV imaging.

Description
The CrystalWand Magnetic is designed to be used exclusively with the CrystalCap Magnetic, CrystalCap Copper Magnetic, and CrystalCap HT systems during transfer of the caps from the vial to the goniometer head and from the goniometer to the vial. The 6 1/2” (165 mm) long chrome plated steel wand features a plastic housing enclosing a spring tensioned plunger that when depressed, moves a non-magnetic steel platform away from the magnet housed in the end of the wand. This causes the steel CrystalCap to detach from the wand and be placed readily into the vial or the CryoTong™. When the platform is retracted, the wand securely holds the steel CrystalCap Magnetic. The CrystalWand Magnetic 45° (McMiken Tool) offers the same functionality but is bent at a 45° angle at approximately 2.5 cm from the magnet end.

The Vial Clamp – Curved is a chrome plated, hemostat style tool. It has a tip shaped to hold the storage vial of all the CrystalCap Systems at an angle when the clamp is closed. The clamp can be locked using the hemostat style closure. The end of the clamp, where the vial is held, is curved at either a 45°/135° or 110°/ 70° angle. When the vial is placed in the clamp, the length of it is positioned to the clamp at either of those same angles. The overall length of each clamp is the same, 195 mm.

Description
The CryoTong is a tool used to manually transfer a crystal mounted on a CrystalCap™ from liquid nitrogen to a magnetic base in a goniometer head positioned in a cryogenic stream, and then back to liquid nitrogen. The one-piece, compact CryoTong is available in two lengths. The long CryoTong is approximately 180 mm in overall length. The artery clamp style maintains the CryoTong in the closed position until the clamp is squeezed, which opens the opposing heads. The heads are non-magnetic stainless steel. The handle is magnetic stainless steel. The inside of the head is machined to closely surround the CrystalCap with loop and crystal in place. A small retaining lip is machined into the lower portion of the head to prevent the CrystalCap from slipping out of the CryoTong when the tool is in the closed position. The CryoTong can maintain the temperature of the crystal at -160°C for up to 30 seconds during room temperature crystal transfers. Choose the proper CryoTong size based upon the pin length required by the configuration of the x-ray data collection hardware used. The 18 mm CryoTong (HR5-112) is to be used with the CrystalCap HT and any other CrystalCap configured for 18 mm.HR5-114 Long CryoTong 18 mm, 180° features the head in alignment with the handle of the CryoTong.

CrystalCap Holders are convenient stands for holding all CrystalCap™ systems in liquid nitrogen dewars during crystal cryogenic procedures. Available in two sizes. Both holders feature spring-clips to secure the CrystalCap Vials in the holder. Both stands have 3 inches of adjustable Z (height) which allows one to raise or lower the CrystalCaps to the appropriate height, depending upon the liquid nitrogen level in the dewar.
